3. Indications for Use.

Endometriosis with an associated infertility, benign neo formations of a lacteal gland (including chronic cystic mastitis), primary menorrhagia, premenstrual syndrome, primary sexual precocity, gynecomastia. Prevention of all the types (cutaneous, abdominal, laryngeal) of aggravations when there is a case of hereditary agioedema.

4. Action

Danazol is a hormonal drug, which suppresses the activity of ovaries, inhibits ovulation, provokes atrophy of endometrium and decreases pain syndrome in women.

5. Posology and Administration

The drug should be taken orally, 2–4 times a day. Administration should be started in the first day of menstrual cycle. When there is endometriosis the starting dose should be 400 mg daily, subsequently it will be up to 800 mg, the course will be 6 months; in the quality of after-surgery preparation it should be taken 300–400 mg daily, 1–2 months before the surgery. In menorrhagias and premenstrual syndrome it will be taken100–400 mg daily – the course should be 3 months. In mastopathy there should be taken 300 mg a day during 3–6 months. In children with sexual precocity a daily dose (from 100 to 400 mg), and the course duration should be chosen strictly individually. For the prevention of angioedema the starting dose should be 200 mg with a succeeded decrease (provided there are no aggravations) during 1–3 months.

6. Counterindications

Sugar diabetes, epilepsy, migraine, disorder of hemostasis plasmatic mechanisms, elderly age.
Hypersensitivity, porphyria (excavation of the disease is possible), cancer of lacteal glands, androgen-dependent tumors, including cancer of prostate, thromboembolia, genital bleedings (before adjustment of their reason), acute renal and/or hepatic failure, manifested heart failure, pregnancy, breastfeeding.

7. Should Be Taken with Precautions, if…

Caution should be taken while curing sufferers of heart and kidneys failure.
During the treatment it is recommended to use non-hormonal methods of contraception.
For sugar diabetes patients an insulin dose correction is needed.

8. Interface with Other Medications

Danazol should not be taken simultaneously with other medications, non-present in the doctor’s order – effect enhancement is possible.

9. Overdosing

Symptoms: headache, dizziness, skin benumbed sensation, “creeping sensation”, convulsions, nausea, vomit.

10. Side Effects

Dizziness, fainting, emotional instability, nervousness, spasm of muscles, headache, increase of intracranial tension, chronic pain in loins, skin benumbed sensation, sleep disorders, vision disorders. Nausea, liver dysfunction. Disorders of menstrual function, among them - irregular and painful periods, also unduly long-lasting ones, moreover, flushes, elevated perspiration, dry vaginal opening, irritation and decrease of the size of lacteal glands. Stuffiness in nose, rash. Heightened danger of bleeding in hemophilia patients.

11. Pregnancy and Lactation

At the onset of pregnancy during Danazol therapy the drug administration should be stopped and the patient should be warned of a possible risk for the fetus.

Regard must be paid in the beginning of the therapy – if side effects do not specially disturb, automobile driving is allowed.
